preparation | preparation of mixture of 1, 2-picolinic acid hydrochloride and 2-picolinic acid methyl ester hydrochloride: 140g of methanol is cooled to 0 ℃, 30g of sulfoxide chloride is added dropwise, 30g of 2-picolinic acid is added after dropwise, gradually heated to 20 ℃ for reaction for 2h, then refluxed for 5h, cooled to 0 ℃, and filtered, after drying at 50 ℃, 38g of white crystal was obtained, HPLC2-methyl picolinate accounted for about 60%, and the yield was higher than 90%. 2. Use the mixture of 2-picolinic acid hydrochloride and 2-picolinic acid methyl ester hydrochloride to prepare 4-chloro-2-picolinic acid methyl ester hydrochloride: add 38g of 2-picolinic acid hydrochloride and 2-picolinic acid methyl ester hydrochloride mixture and 3g of sodium bromide to 150g of thionyl chloride, slowly heat up to reflux, and then keep reflux for 16h. After chlorination is completed, the temperature is lowered to 40 ℃, 200g of toluene is added, sulfoxide chloride is evaporated under reduced pressure, the temperature is lowered to 10 ℃, 50g of methanol is added dropwise and refluxed for 2h. Then cool down and evaporate the methanol under reduced pressure, then cool down to 0 ℃, filter, filter cake washed with toluene, suspend in water at 10 ℃, add sodium carbonate, and precipitate light yellow solid. The solid was recrystallized with water after suction filtration and dried at room temperature for 18h to obtain 25g of white needle crystal. |
